Advanced Emulsification: Mastering High-Viscosity Ointments with a 30L GMP Mixer

In pharmaceutical manufacturing, creating a stable, elegant, and efficacious topical ointment is a significant technical challenge. Ointments are often formulated with a high percentage of active pharmaceutical ingredients (APIs), thickeners, and fillers, resulting in a highly viscous paste that is difficult to mix and deaerate effectively. The primary challenge in processing high-viscosity ointments is achieving a uniform dispersion and a fine particle size. Standard mixers struggle with thick pastes, creating "dead zones" where unmixed material accumulates. The 30L mixer overcomes this with its powerful, bottom-mounted homogenizer. As the material is thick and paste-like, gravity naturally feeds it down into the high-shear zone of the rotor-stator assembly. This is a key advantage over top-mounted homogenizers, which rely on creating a vortex that high-viscosity materials cannot sustain. The rotor-stator, spinning at up to 3000 rpm, generates intense mechanical and hydraulic shear, breaking down agglomerates and reducing the API and excipient particles to the 2-5 micron range. To complement the homogenizer, the slow-speed scraper agitator plays an equally vital role. Featuring PTFE blades, it continuously sweeps the tank walls, preventing the ointment from sticking and burning or degrading due to localized overheating. This wall-scraping action also continuously returns the material to the center of the tank, where it is fed into the homogenizer, ensuring that the entire batch, and not just the center, is processed uniformly. This combination of bottom shearing and wall scraping is essential for achieving homogeneity in 100% of the batch, which is a strict requirement for pharmaceutical products.

The vacuum system is the third pillar of the 30L mixer's success with viscous materials. During the high-shear mixing of thick pastes, a significant amount of air is inevitably incorporated, creating bubbles. These bubbles can compromise the product's appearance, create sites for microbial growth, and affect dosage accuracy. The vacuum environment, operating at -0.09 MPa, pulls out all entrapped air, resulting in a dense, bubble-free, and visually perfect ointment.
